RESUMO

Purpose To synthesize and characterize poly(hydroxybutyrate) (PHB) and norbixin membranes to evaluate them for genotoxicity in rats and wound healing in mice by histological staining. Methods For the evaluation of genotoxicity, male rats ( Rattus novegicus ) were divided into three groups (n= 5): 5% PHB/Norbixin membrane introduced into the peritoneum by laparotomy; B - negative control; C - positive control (intraperitoneal dose of cyclophosphamide 50 mg/kg). For the evaluation of biocompatibilty, a cutaneous wound was induced on the back of males mice ( Mus musculus ) divided into two experimental treatment groups: control and membrane that underwent euthanasia after 7 and 14 days treatment. Statistical analysis ware made by One Way Anova post hoc Tukey Test (p<0.05). Results Regarding the incidence of polychromatic erythrocytes, there was no difference between negative control and 5% PHB/Norbixin membrane; however, when compared to the positive control represented by cyclophosphamide, there was a significant difference (p <0.001). As for DNA damage, the changes induced in the first 4h were repaired in 24h. In addition, the membrane was effective in abbreviating the inflammatory process and served as a scaffold due to the stimulus to reepithelialization mainly on the 7 days of treatment. Conclusion The non-genotoxic PHB/Norbixin 5% membrane presented promising results that suggest its effectiveness as a guide for tissue regeneration given its biocompatibility.

PURPOSE: To evaluate the in vivo response of photobiomodulation therapy associated with norbixin-based poly(hydroxybutyrate) membrane (PHB) in tenotomized calcaneal tendon. METHODS: Thirty rats were randomly allocated to six groups (n=5 each): LED groups (L1, L2 and L3) and membrane + LED groups (ML1, ML2 and ML3). The right calcaneal tendons of all animals were sectioned transversely and were irradiated with LED daily, one hour after surgery every 24 hours, until the day of euthanasia. At the end of the experiments the tendons were removed for histological analysis. RESULTS: The histological analysis showed a significant reduction in inflammatory cells in the ML1, ML2 and ML3 groups (p=0.0056, p=0.0018 and p<0.0001, respectively) compared to those in the LED group. There was greater proliferation of fibroblasts in the ML1 (p<0.0001) and L3 (p<0.0001) groups. A higher concentration of type I collagen was also observed in the ML1 group (p=0.0043) replacing type III collagen. CONCLUSION: Photobiomodulation in association with norbixin-based PHB membrane led to control of the inflammatory process. However, it did not favor fibroblast proliferation and did not optimize type I collagen formation in the expected stage of the repair process.

A pesquisa teve como objetivo comparar os resultados dos métodos clássicos de umidade e resíduo mineral (cinzas) com os resultados encontrados pela termogravimetria (TG) e derivada da termogravimetria (DTG) na amostra de farinha de jenipapo desidratada em estufa (FJDE). Utilizaram-se os testes de umidade e cinzas pelo método convencional e pela termogravimetria (TG) e derivada de termogravemetria (DTG). Os resultados obtidos foram 7,86 ± 0,3 % para umidade convencional e 7,774 % para umidade na TG, cinzas convencional 3,38 ± 0,025% e cinzas por TG com 4,4 %. Conclui-se que as análises termogravimétricas apresentaram resultados próximos aos modelos convencionais para a umidade e cinzas, sendo que é realizada em um menor espaço de tempo, sem haver manipulação da amostra pelo analista no meio do processo evitando distorções nos resultados.

Biocompatible membranes are widely used in medicine to stimulate bone repair. Several studies have demonstrated that laser photobiomodulation (PBM) also stimulates osteoblast proliferation and osteogenesis at the fracture site, leading to a greater deposition of bone mass and accelerating the process of bone consolidation. This work assessed the therapeutic effect of 780-nm laser PBM and a polystyrene membrane coated with norbixin and collagen (PSNC) on bone healing in rats with calvarial bone defect. Histological staining, Raman spectroscopy, and scanning electron microscopy (SEM) were used to evaluate the bone repair process. Four experimental treatment groups were compared: C, control; M, membrane only; L, laser PBM only; and ML, membrane + laser PBM. A bone defect was created in the calvaria of each animal, with each group subdivided into two subgroups that underwent euthanasia after 15 and 30 days treatment. The L and ML groups were irradiated (λ = 780 nm, ED = 6 J/cm2, P = 60 mW, t = 4 s) postoperatively on alternate days until they were euthanized. The bone concentration of hydroxyapatite (CHA) showed a clear gradation with increasing phosphate area in the order B (normal cortical bone) > L > M > ML > C for both periods. The PSNC membrane was effective in reducing the inflammatory process and served as a scaffold for bone repair. The laser PBM also showed positive effects on the bone repair process with increased deposition and organization of the newly formed bone. However, laser PBM failed to improve the bioactive properties of the membrane scaffold.

Introdução: A queimadura é uma injúria que altera a integridade da pele, decorrente de traumas térmicos. Dentre os compostos não convencionais utilizados na cicatrização de feridas estão a norbixina e as preparações com prata. Objetivo: avaliar a ação cicatrizante da nanopartícula de prata associada à norbixina em queimaduras de Mus musculus. Métodos: Inicialmente, preparou-se uma solução nanoprata/norbixina e a partir desta obteve-se o gel teste. Posteriormente dividiu-se os animais em 3 grupos, sendo negativo (sem tratamento), positivo (sulfadiazina de prata) e teste (gel teste). Nestes animais houve a indução das queimaduras. No 7º e 14º dia realizou-se a eutanásia e os tecidos lesionados foram coletados para análise microscópica. Resultado: Verificou-se potencial vantagem do grupo teste em relação ao negativo e igualdade comparada ao positivo. Conclusão: A nanopartícula de prata associada à norbixina representa uma fonte alternativa para a cicatrização de queimaduras. (AU)


Introduction: Burning is an injury that alters the integrity of the skin, due to thermal trauma. Among the non-conventional compounds used in wound healing are norbixin and silver preparations. Objective: to evaluate the cicatrizing action of silver nanoparticle associated with norbixina in Mus musculus burns. Methods: Initially, a nanoprata / norbixin solution was prepared and from this solution the test gel was obtained. Afterwards the animals were divided into 3 groups, being negative (without treatment), positive (silver sulfadiazine) and test (gel test). In these animals, there was the induction of burns. On the 7th and 14th day, euthanasia was performed and the injured tissues were collected for microscopic analysis. Result: There was a potential advantage of the test group in relation to negative and equality compared to positive. Conclusion: The silver nanoparticle associated with norbixin represents an alternative source for the healing of burns. (AU)

Objetivo: detectar possíveis alterações em órgãos de ratos Wistar (Rattus norvegicus) após a administraçãopor via oral de prata coloidal para posterior análise histológica. Metodologia: este estudo experimental comabordagem quanti-qualitativa, consistiu em 24 ratas não-isogênicas, nulíparas, com massa corpórea entre de190 a 260 gramas, divididas aleatoriamente em dois grupos, controle e experimento. Os ratos receberamdoses diárias de 5 mL/kg de prata coloidal. Resultados: no grupo controle não se detectou quaisqueralterações orgânicas. O grupo experimento, porém, apresentou alterações renais e hepáticas após o 14oeentre o 7oe 14odia de exposição, respectivamente. Discussão: a análise histopatológica dos rins dos animaisevidenciou que todos os 12 animais do grupo tratado com prata coloidal apresentaram nefrite, congestão,tumefação turva e espessamento de alças glomerulares. As alterações hepáticas foram congestão e tumefaçãoturva, núcleo aumentado e vacuolizado. Conclusão: os achados apontam potencial toxicidade da pratacoloidal aos rins e fígados de ratas Wistar.


Objective: to detect possible alterations in Wistar rats’ (Rattus norvegicus) organs after oral administrationof colloidal silver for subsequent histological analysis. Methodology: this experimental study with quantitativeand qualitative approach consisted of 24 non-isogenic, nulliparous rats, with body mass between 190-260grams, randomly divided into two groups, control and experiment. The rats received daily doses of 10 mL/kgof colloidal silver. Results: in the control group, no organic alterations were detected. The experimentalgroup, however, presented kidney and liver alterations after the 14th and between the 7th and 14th day ofexposure, respectively. Discussion: the histopathological analysis of the kidneys of these animals showed thatall twelve treated with colloidal silver showed nephritis, congestion, cloudy swelling, and thickening ofglomerular loops. Hepatic lesions were congestion and cloudy swelling, and increased and vacuolated nucleus.Conclusion: the findings indicated potential toxicity of colloidal silver to the kidneys and liver of Wistar rats.

Objetivo: descrever o uso de alimentos e suplementos alimentares com atividade antioxidante em idosos frequentadores de dois centros de convivência. Metodologia: Pesquisa descritiva de abordagem quantitativa, desenvolvida no âmbito do Programa Institucional de Bolsas de Iniciação Científica da Faculdade Integral Diferencial, cujo objetivo foi descrever o uso de alimentos e suplementos alimentares com atividade antioxidante em idosos. A amostra constituiu-se de 152 sujeitos com idade igual ou superior a 60 anos e frequentadores ativos em dois Centros de Convivência da Terceira Idade no município de Teresina-Piauí, que voluntariamente se dispuseram a aceitar responder ao Termo de Consentimento Livre e Esclarecido. Utilizou-se como instrumento para coleta de dados um questionário de entrevista que abordou questões relacionadas ao conhecimento, frequência, quantidade e qualidade do consumo de frutas, legumes e verduras e suplementação alimentar. Resultados: todos os idosos pesquisados consomem algum tipo de alimento com propriedade antioxidante, predominando, majoritariamente: compostos fenólicos, carotenoides e vitamina C, com variação de frequência e, em 77,6% dos casos, associada ao uso de suplementação alimentar. Os mesmos possuem pouco conhecimento acerca da propriedade antioxidante dos alimentos. Conclusão: A grande maioria considera importante a inclusão destes na dieta. A prevalência do uso da suplementação alimentar é elevada.


Objective: To describe the use of foods and dietary supplements with antioxidant activity in elderly patrons of two centers. Methods: Descriptive study of quantitative approach, developed within the Institutional Program for Scientific Initiation Scholarships School of Integral Differential, whose aim was to describe the use of foods and dietary supplements with antioxidant activity in the elderly. The sample consisted of 152 subjects aged over 60 years and active patrons in two Centres for the Elderly Living in the city of Teresina, Piauí, who willingly volunteered to accept answer the Statement of Informed Consent. Was used as an instrument for data collection a questionnaire interview that addressed issues related to knowledge, frequency, quantity and quality of fruit, vegetables and food supplementation. Results: all seniors surveyed consume any type of food with antioxidant properties, predominantly, overwhelmingly, phenolic compounds, carotenoids, and vitamin C, with varying frequency and in 77.6% of cases associated with the use of dietary supplementation. They have little knowledge about the antioxidant properties of foods. Conclusion: The majority considers the inclusion of these in the diet. The prevalence of use of dietary supplementation is high.
